In our journey to understand the cause of the pain and, in particular, migraine headaches, we studied in depth for an answer. Now trustfully claim that we can cure but do not treat or abort migraine headaches. In our terms, cure means relief from migraine for years coming without the use of any drug. We understand that Pain, particularly migraine headaches, is part of the cosmic consciousness on Earth. Happiness and Pain are twins. Happiness is absent whenever our body is in Pain, and vice versa. Pain is a characteristic alignment of perishable creatures programmed by DNA on the blue planet, the Earth; in contrast, physical elements of the physical world and the surrounding universe survive a long existence beyond our life. An upregulated hypersensitivity of the parasympathetic reflex to repetitive physical or chemical stressors disrupts secure equilibrium, the integrity of the brain, and its reflective sensory organs. A sophisticated signaling system translates this immunologic reaction into the eruption of neurotoxins such as TNF, Nitric Oxide, Interleukins, and other known neurotoxins. We speculate that primary migraine headaches are based on an immunologic process in response to physical elements of the planet Earth, the surrounding cosmic invisible electromagnetic rays, and the extra forces of the physical nature of our metaphysic. Primary migraine seems to be frequently reactivated by repeated confrontation with the physical forces surrounding us called triggers. Physical insults vary in nature and quality, internal and external sources. For instance, an aneurysm or tumor in the brain is a pressure and physical source. An insult to the brain initiates and provokes a series of cascades of complex immunologic neuroexcitation involving neurotoxins and proteins-peptides to relay the unfriendly assault to ganglions and certain centers of the brain for action. In molecular physiologic terms, we face a neurogenic hypersensitivity chain reaction—a “Neuro-Allergy” to the forces of physics! Once the resolution of the stimulus is achieved, the degradation of the receptor and TNF-α, non-canonical NF-κB complex proceeds. Upregulated hypersensitivity of the neurons proceeds with the degradation signaling of the active complex.

Understanding of the NF-κB (nuclear factor-κB) activation signaling pathways is the key to the biological function of the pathway in immunity and inflammation. [Shao-Cong Sun – Nature Reviews Immunology, volume 17, pages 545–558 (2017)]. NF-κB is activated as a first-line response of vital cells to internal and external stimuli, which are not compatible with the vitality of cells and tissue. The biological function of NF-κB is regulated by two important pathways: canonical and non-canonical signaling systems. Free radicals, viral antigens, bacteria, heavy metals, cytokines, stress, visible and invisible electromagnetic irradiation and rays, and other components of cosmic influences in human and almost all living species activate the biological NF-κB signaling [Shao-Cong Sun – Nature Reviews Immunology].

It is well established that canonical signaling of the induced TNFRs (Tumor Necrosing Factor Receptors), activation, and clonal expansion activate non-canonical NF-κB signaling. Different cell and tissue types are associated with different responses to autoimmunity and inflammation. Inflammation is a response to an immune signaling system promoting awareness for an appropriate reaction. We suggest that the migraine headache mechanism and process is a neurovascular dynamic. Vaso-constriction, but not vasodilation, in the property of peripheral sensory nerves is the initiator of the signaling of neurotoxin synthesis. The regulation of this process is translated by incredible DNA encoding in the autonomous nervous system. A reactive response to potentially destructive vasoconstriction and hypoxia is vasodilation through upregulating break signals and spillover of the Calcium Gene Receptor Peptides in the properties of the CNS, peripheral ganglia, and peripheral nerves. The brain, per se, does not carry pain receptors. We understand that repetitive signaling for chronic migraine headaches is related to disbalance in the sympathetic and parasympathetic nerve system, interrupting their equilibrium. The disbalance kinetic upregulation toward hypersensitivity of the parasympathetic response by neurotoxin cascades implemented in the process. Our treatment suggested regulating the stability of the properties of the sympathetic and parasympathetic signaling systems, desensitizing the primarily hypersensitized sensory organ. We understand that repeated exposure to the same physical energy and stimulus would trigger the process of migraine headaches even after years of being in silence. Auras of any type are concomitant, signaling varieties of sensory organs, particularly the eyes. The upregulation of the proinflammatory toxins could be interrupted by certain anti-inflammatory drugs (NSAIDs), the Triptan family of drugs, and selective serotonin receptor agonists. Even herbal supplements, darkness, and electrostimulation of the sensory nerves in the periphery essentially show temporary interruption of the autonomous nerves, providing short-term relief, but not a cure or long-term relief.

Molecular biological effect of glucocorticoids by penetrating the cell membrane and binding to DNA induces suppression of transcription by opposing transcription factors AP-1 and NF-κB. AP-1 and TNF-α, which potentiate genes encoding almost all proinflammatory cytokines. (5,6,7,8,9,10) *

Glucocorticoids also suppress expression of inflammatory genes encoding T cell growth factors such as IL-2, IL-4, IL-15, and IL-17 as well as interferon-γ (IFN-γ). In addition, glucocorticoids reduce expression of genes encoding COX-2, inducible nitric oxide synthase, and intracellular adhesion molecule-1 (ICAM-1), which are normally induced by the cytokines IL-1β and TNF-α. Glucocorticoids increase expression of genes encoding anti-inflammatory molecules, such as the cytokine IL-10 and the IL-1 type 2 decoy receptor. (11) *.

The gene suppressive effect of glucocorticoids, per se, Dexamethasone switches the key signals by silencing the hyper-activated proinflammatory signals. The promotion of those signals may encode a balance between sympathetic/parasympathetic mechanisms at the level of vascular-neuronal capillaries. Induction of nitric oxide synthase, changing the status of hypoxia, consequently relieves pain by the inhibition of COX-2 and inhibition of nearly all proinflammatory cytokine genes (19,23,24,25) * that play an important role in the pain signaling.
